Colchester United turned the tables on Cheltenham Town for their first round LDV Vans Trophy defeat at Whaddon Road last season.
The Robins won that encounter 4-1 and looked on course for another victory when Martin Devaney gave them a 17th minute lead after a pass from John Finnigan had fallen kindly for him off the head of defender Scott Fitzgerald.
Second Division Colchester displayed that little bit more quality after that and drew level five minutes before the break when Craig Fagan supplied Joe Keith and the midfielder scored with a low angled shot.
Cheltenham's Ben Cleverley hit the post almost straightaway and the home side felt they should have had a penalty when veteran striker Bob Taylor went to ground under a challenge from goalkeeper Simon Brown but referee Steve Tanner was unmoved.
Colchester cruised past their hosts in the 65th minute when a poor backpass by Kayode Odejayi was intercepted by substitute Rowan Vine and the on-loan Portsmouth player rounded goalkeeper Shane Higgs to score.
Cheltenham substitute Damian Spencer missed a couple of late chances to level but Vine completed the win two minutes from time with a header from a Thomas Pinault cross.
